Charlo defeated Cimmaron Davis in his professional debut on August 12, 2008 by TKO in the second round. On July 11, 2009, Charlo faced Deon Nash, whom his brother Jermell had previously fought and defeated in 2008.

After defeating Nash by unanimous decision, he challenged Nash to a rematch on August 28, 2010, which Charlo eventually won via corner submission.
