A man has been killed when his car was struck by a vehicle that had driven away from a police stop at high speed in western Sydney on Tuesday morning.

Motorists are being diverted due to the closure of the Great Western Highway in both directions at St Marys, between Glossop and Sydney streets.

A marked police car stopped near a Holden Insignia sedan parked on Mamre Road at St Marys at about 4am.

The driver then allegedly accelerated away towards the Great Western Highway and the police followed but lost sight of the car.

The Holden then hit a Toyota Corolla hatchback, killing the male driver who was pronounced dead at the scene.

The man has not yet been formally identified but is believed to be aged in his 40s.

The Holden driver allegedly fled from the scene on foot.

A crime scene has been established and a critical incident team from North Shore Police Area Command was investigating.

The investigation is also subject to an independent review.

Anyone with dashcam footage, CCTV vision or information about the incident is urged to contact police or Crime Stoppers on 1800 333 000.

Diversions are in place, with light vehicles being directed to Glossop, Brisbane and Sydney streets in both directions.

Heavy vehicles can use Mamre Road, the M4 and the M7.
